COMMENTARY Wound repair, keratinocyte activation and integrin modulation

چکیده
Modern cell biology is on the verge of answering Weiss's question. Recent studies have identified the adhesion systems that participate in 'bobbins' (i.e., hemidesmosomes) and the changes in adhesion receptors that can account for keratinocyte activation, that is, the transition of keratinocytes from a sedentary to migratory lifestyle. In this commentary, I will review some of the recent research on changes that occur during activation of normal keratinocytes to wound keratinocytes and discuss the role of integrin receptor modulation in these changes.

α6ß4 integrin regulates keratinocyte chemotaxis through differential GTPase activation and antagonism of α3ß1 integrin
Growth factor-induced cell migration and proliferation are essential for epithelial wound repair. Cell migration during wound repair also depends upon expression of laminin-5, a ligand for α6ß4 integrin. We investigated the role of α6ß4 integrin in laminin-5-dependent keratinocyte migration by re-expressing normal or attachment-defective ß4 integrin in ß4 integrin null keratinocytes. We found t...
متن کاملCrucial role of vinexin for keratinocyte migration in vitro and epidermal wound healing in vivo.
In the process of tissue injury and repair, epithelial cells rapidly migrate and form epithelial sheets. Vinexin is a cytoplasmic molecule of the integrin-containing cell adhesion complex localized at focal contacts in vitro. Here, we investigated the roles of vinexin in keratinocyte migration in vitro and wound healing in vivo. Vinexin knockdown using siRNA delayed migration of both HaCaT huma...
متن کاملkeratinocyte polarity through integrin - mediated modulation of Rac 1 and cofilin activities
integrin, a component of hemidesmosomes, also plays a role in keratinocyte migration via signaling through Rac1 to the actin-severing protein cofilin. Here, we tested the hypothesis that the 4 integrin-associated plakin protein, bullous pemphigoid antigen 1e (BPAG1e) functions as a scaffold for Rac1/ cofilin signal transduction. We generated keratinocyte lines exhibiting a stable knockdow...
متن کاملActivation of keratinocyte fibronectin receptor function during cutaneous wound healing.
Keratinocytes freshly isolated from unwounded skin could not attach and spread on fibronectin (FN)-coated culture dishes and could not bind and phagocytose FN-coated beads. These adhesive functions were activated, however, in keratinocytes that were isolated from healing wounds. Moreover, adhesiveness of basal keratinocytes to FN substrata was activated during epidermal cell or explant culture....
متن کاملIntegrin a3b1-Dependent Activation of FAK/Src Regulates Rac1-Mediated Keratinocyte Polarization on Laminin-5
Upon epidermal wounding, keratinocytes at the wound edge become activated, deposit newly synthesized laminin-5 into the extracellular matrix, and migrate into the wound bed. The interaction between integrin a3b1 and laminin-5 is essential for establishment of a stable, leading lamellipodium and persistent keratinocyte migration. We previously showed that integrin a3b1 activates the Rho family G...
